تعیین روابط فیلوژنتیک تعدادی از ژنوتیپ‌های مرکبات شمال ایران با استفاده از نشانگرهای ISSR

چکیده

اطلاع از روابط فیلوژنتیک و تنوع ژنتیکی در مرکبات جهت تشخیص روابط ژنتیکی، شناسایی ژرم‌پلاسم و ثبت ارقام جدید، امری مهم و ضروری می‌باشد. در کلکسیون‌های مرکبات کشور، تعدادی بیوتیپ‌ وجود دارد که شناسایی آنها عمدتاً براساس صفات مورفولوژی بوده و تحقیقات ژنتیکی چندانی بر روی آنها انجام نشده است به­منظور بررسی تنوع ژنتیکی و روابط خویشاوندی بین 45 ژنوتیپ از ارقام ناشناخته و تجاری مرکبات موجود در ایستگاه تحقیقات کترا، از نشانگرهای ISSR استفاده گردید. دندروگرام حاصل از تجزیه خوشه‌ای به‌ روش UPGMA با ضریب تشابه جاکارد ژنوتیپ‌های مورد بررسی را به نه گروه (A، B، C، D، E، F، G، H و I) مجزا طبقه‌بندی کرد. ارقام پرتقال، نارنگی و 19 ژنوتیپ ناشناخته در گروه A، یک ژنوتیپ ناشناخته در گروه B، دو ژنوتیپ ناشناخته در گروه C، یک ژنوتیپ ناشناخته در گروه D، گریپ‌فروت و پوملو و نارنج و شش ژنوتیپ ناشناخته در گروه E، دو ژنوتیپ ناشناخته در گروهF ، یوزو در گروه G، دو ژنوتیپ ناشناخته در گروه H و در نهایت ارقام لیمو و بالنگ در گروه I قرار گرفتند. تجزیه تنوع ژنتیکی و تعیین روابط خویشاوندی در مرکبات، اطلاعات مفیدی را برای برنامه‌های به‌نژادی، انتخاب و حفظ ارقام فراهم می‌کند.

Understanding phylogenic relationships and genetic diversity in Citrusis considered to be important in clarifying their genetic relationships, germplasm characterization and the registration of new varieties. There are some Citrus accessions in the country Citrus collections which have been classified merely based on their morphological traits. Molecular markers would help to infer their relationships with known cultivars. In the present research work, phylogenic relationships among 45 Citrus genotypes, including 33 unknown local genotypes and 12 commercially important Citrus varieties at the Collection in agricultural research institute of Kotra were investigated through ISSR markers. Genetic similarities among accessions were calculated according to the Jaccard Similarity Index and used to construct a dendrogram based on the unweighted pair groups method arithmetic averages (UPGMA), which put the 45 samples into nine major groups. Orange, mandarin and 19 unknown genotypes into group A, an unknown genotype into group B, two unknown genotypes into group C, an unknown genotype into group  D, grapefruit, Pummelo, sour orange and six unknown genotypes into group E, two unknown genotypes into group F, Yuzo into group G, two unknown genotypes into group H and citron, Eureka lemon, sweet lime, Mexican lime  into group I. Genetic diversity and phylogenetic analysis in citrus, provide useful information for further breeding programs, collection, preservation and utilization.
