As its name implies, lipid transfer proteins are responsible for the shuttling of phospholipids and other fatty acid groups between cell membranes. Here we study the expression pattern of Lipid transfer protein gene from wheat (Triticum aestivum) responding to Septoria tritici Blotch (STB) by semi quantitative reverse transcriptase-polymerase chain reaction (RT-PCR). This method is used to qualitatively detect gene expression in different cells. level of LTP2 gene expression was measured over time, from 0h to 6 days after inoculation in Wangshuibai as a resistant wheat cultivar and Falat as a sensitive one. The results showed that inoculation of wheat by M. graminicola (asexual stage: Septoria tritici) cause different level of LTP2 gene expression in both resistant and susceptible cultivars over time. The pattern of gene expression indicated that LTP2 gene is an early expression gene in resistant cultivar and induced at 24h after inoculation and in sensitive cultivare the high level expression have been seen at 6 days after inoculation. Thus, according this results can be concluded that these genes, along the main genes, increase and maintain resistance to Septoria tritici blotch in wheat. In this study we cloned LTP2 genes from both cultivars and after sequencing the blast protein was studied. Blast results confirmed that these proteins contain eight cysteines that are conserved in all LTP peptides.}, keywords = {Mycosphaerella graminicola,Expression of gene LTP2,Triticum aestivum}, title_fa = {بررسی الگوی بیان ژن کدکننده پروتئین انتقال‌دهنده چربی در مقابله با بیماری سپتوریوز برگی در گندم نان و کلون‌سازی آن}, abstract_fa = {پروتئین‌های انتقال‌دهنده چربی (LTP)، در حدود 10-7 کیلودالتون وزن داشته و به‌دلیل توانایی که در انتقال انواع مولکول‌های فسفولیپید از محل سنتز (شبکه آندوپلاسمی) به غشاهای سلولی دارند، به این نام خوانده می‌شوند. در این مطالعه نقش این پروتئین‌ها در ایجاد مقاومت گندم نان نسبت به بیماری سپتوریوز برگی (Septoria tritici) با بررسی الگوی بیان ژنLTP2  طی 6-0 روز بعد از آلودگی در رقم مقاوم ونگشوبایی و حساس فلات با استفاده از روشRT-PCR  نیمه­کمی بررسی گردید. از آن‌جا که این روش امکان مقایسه نسبی مقدار رونوشت ژن‌ها را در انواع سلول‌های مختلف فراهم می‌نماید مطالعه‌ی الگوی بیان ژن به سادگی ممکن می‌گردد. که طی آن علاوه‌بر میزان بیان، زمان بیان ژن نیز مورد بررسی قرار می‌گیرد. نتایج این تحقیق نشان داد که میزان بیان ژن در اثر آلودگی گیاه با بیماری سپتوریوز برگی در هر دو رقم حساس و مقاوم افزایش می‌یابد. بیان این ژن در رقم مقاوم طی 24 ساعت بعد از آلودگی افزایش می‌یابد در‌حالی‌که این افزایش بیان در رقم حساس با تأخیر و بعد از شش روز مشاهده می‌شود. با توجه به نتایج حاصل می‌توان گفت این ژن با ایجاد مقاومت علیه بیماری سپتوریوز برگی مرتبط بوده و در کنار ژن‌های اصلی ایجادکننده مقاومت باعث تشدید و حفظ مقاومت خواهد شد. هم‌چنین ژن پس از کلون‌سازی، تعیین توالی گردید و مورد بررسی بلاست پروتئینی قرار گرفت. با بررسی نتایج بلاست مشخص گشت که توالی کلون شده دارای هشت سیستئین ثابت و حفاظت شده است که در همه پروتئین‌های انتقال‌دهنده چربی ثابت می‌باشد.}, keywords_fa = {Mycosphaerella graminicola,تظاهر ژن LTP2,Triticum aestivum}, url = {https://ab.basu.ac.ir/article_2268.html}, eprint = {https://ab.basu.ac.ir/article_2268_bc2e2478c49ae5920bdb09f268ebd782.pdf} }
