This experiment has taken place in Golestan province Agriculture research station in split plot format with RCBD design with four replicates. Drought stress as a main factor included three levels (irrigation after 50, 100, and 150 millimeters evaporation from the A class evaporation pot) of which 50 millimeters evaporation is considered as control. The sub factor included factorial combination of 3 varieties (DPX, Sari and WE6) and three sampling stages (flowering, packing pod and grain filling). Evaluated traits included A and B chlorophyll amount and alternation in gene expression. The amount of chlorophyll was increased on first step of the stress condition (100 ml evaporation) and decreased again, on the second step (150 ml evaporation). The TBARM level was increased under the stress condition as well as growth stages. The gene expression analysis was carried out by using QRT-PCR technique. The results showed at first level of drought stress condition catalase gene expression had the most amounts. It was quite similar to DREB1 gene expression. DPX variety showed the most DREB1 gene expression first level of drought stress condition in packing pod stage, and least reduction in second level of the drought stress condition. Sari variety showed the most reduction in DREB1 gene expression in second level of the drought stress condition. Chlorophyll amount also showed a positive correlation with the catalase and DREB1 genes expression.}, keywords = {Gene expression,Drought stress,Soybean,Catalase,Chlorophyll amount,DREB1}, title_fa = {ارزیابی تغییرات میزان کلروفیل و بیان ژن‌های کاتالاز و DREB1 در ارقام سویا در شرایط تنش خشکی}, abstract_fa = {تنش خشکی یکی از عوامل مهم محدودکننده تولید محصولات زراعی در مناطق گرمسیری و نیمه­گرمسیری در سراسر جهان است. این آزمایش در ایستگاه تحقیقاتی مرکز تحقیقات کشاورزی استان گلستان و به­صورت اسپلیت­پلات فاکتوریل با طرح پایه بلوک کامل تصادفی در چهار تکرار انجام شد. تنش خشکی به­عنوان فاکتور اصلی شامل سه سطح (آبیاری بعد از 50، 100 و 150 میلی­متر تبخیر از سطح تشتک تبخیر کلاس A) بود که 50 میلی­متر تبخیر به­عنوان شاهد در نظر گرفته شد. ترکیب فاکتوریل سه رقم سویا شامل DPX، ساری و WE6، و سه مرحله رشد (گلدهی، غلاف­بندی و پرشدن دانه) به­عنوان فاکتور فرعی در نظر گرفته شدند. صفات مورد ارزیابی شامل میزان کلروفیل a و b و روند بیان ژن‌های کاتالاز و DREB1 بود. میزان کلروفیل در سطح اول تنش (100 میلی­متر تبخیر) افزایش و در سطح دوم (150 میلی­متر تبخیر) تنش کاهش یافت. بررسی مولکولی بر روی الگوی بیان ژن کاتالاز و DREB1 با استفاده از روش QRT-PCR نشان داد که در طول تنش میزان بیان ژن کاتالاز ابتدا افزایش و سپس کاهش داشت. بیان ژن DREB1 نیز مشابه با ژن کاتالاز بود. رقم DPX بیش­ترین بیان ژن DREB1 را در سطح اول تنش در مرحله­ی غلاف­بندی و کم‌ترین کاهش را در سطح دوم تنش در همین مرحله نشان داد. رقم ساری بیشترین کاهش را در بیان ژن DREB1 در سطح دوم تنش نشان داد. میزان کلروفیل نیز با بیان ژن‌های کاتالاز و DREB1 همبستگی مثبت نشان داد.}, keywords_fa = {بیان ژن,تنش خشکی,سویا,کاتالاز,میزان کلروفیل,DREB1}, url = {https://ab.basu.ac.ir/article_902.html}, eprint = {https://ab.basu.ac.ir/article_902_314a86563efd786f68699caf21caf9ee.pdf} }
