مطالعهی جنینزایی سوماتیکی و تولید بذر مصنوعی از ارقام مختلف انگور (.Vitis vinifera L)
تکثیر ارقام بیدانه انگور به روش بذر مصنوعی دارای اهمیت میباشد. بهمنظور بررسی تأثیر ارقام مختلف انگور و محیطکشت بر جنینزایی سوماتیکی و تولید بذر مصنوعی، آزمایشی بهصورت فاکتوریل بر پایه طرح کاملاً تصادفی با چهار تکرار انجام شد. ریزنمونه برگ چهار رقم انگور در محیط پایهMS 2/1، و در چهار سطح مختلف تنظیمکنندهای رشد کشت داده شد. نتایج نشان داد که تفاوت بسیار معنیداری میان ارقام و سطوح تنظیمکننده رشد بر روی جنینزایی سوماتیکی وجود دارد؛ اما اثر متقابل آنها در سطح (5 درصد) معنیدار نشد. سطح تنظیمکننده رشد (mg/l NAA 2/0 + mg/l TDZ 8/3) بیشترین جنینزایی (65 درصد) را نشان داد. همچنین اثر دو ترکیب هیدروژل کپسول بذر مصنوعی ](محیط کامل نمکهای ماکرو B5 و نمکهای میکرو محیط MS بهعنوان محیط اول کپسول)، (محیط 2/1 نمکهای ماکرو محیط B5 و نمکهای میکرو محیط MS بهعنوان محیط دوم کپسول)[ و چهار رقم انگور بر میزان جوانهزنی بذور، موردبررسی قرار گرفت. نتایج نشان داد که میان ارقام تفاوت معنیداری و میان دو محیط ترکیب کپسوله تفاوت بسیار معنیدار (سطح 1 درصد) وجود دارد. در میان ارقام، رقم کریمسون سیدلس بیشترین بذر جوانهزده شده (65 درصد) را در تیمار محیط دوم کپسوله داشت. مقایسه میانگین جوانهزنی دو محیط کپسوله نشان داد که محیط اول کپسوله (62/52 درصد جوانهزنی) نسبت به محیط دوم کپسوله (62/20 درصد جوانهزنی) برای کپسوله کردن بذور مصنوعی ارقام انگور موجود در این بررسی مناسب است.

اثر نانوذرات دیاکسید تیتانیوم (TiO2)، نور و ساکارز بر جوانهزنی و رشد دانهال توت سیاه (.Morus nigra L) در شرایط درونشیشهای
توت سیاه بومی ایران بوده به لحاظ تغذیهای و فضای سبز دارای اهمیت فراوانی میباشد. با هدف بهینه سازی تکثیر توت سیاه توسط بذر در شرایط کنترل شده، سه آزمایش مجزا در قالب طرح کاملاً تصادفی در پنج تکرار در آزمایشگاه کشت بافت گروه باغبانی دانشگاه شهید چمران اهواز انجام شد. آزمایش اول شامل دو تیمار تاریکی مطلق و شرایط نوری 16 ساعت روشنایی و 8 ساعت تاریکی، آزمایش دوم شامل بررسی اثر غلظتهای مختلف ساکارز (0 (صفر)، 15، 20 و 30 گرم بر لیتر) و آزمایش سوم شامل کاشت بذور بر روی محیطکشت MS کامل محتوی 15 گرم بر لیتر ساکارز و غلظتهای مختلف نانوذرات اکسید تیتانیوم (0 (صفر)، 2/0، 4/0، 6/0 و 1 میلیگرم بر میلیلیتر) در شرایط 16 ساعت روشنایی بود. نتایج آزمایش اول نشان داد که 16 ساعت روشنایی و 8 ساعت تاریکی تأثیر بهتری بر شاخصهای جوانهزنی توت سیاه دارد. نتایج آزمایش دوم نشان داد که غلظت 15 گرم بر لیتر ساکارز نسبت به سایر غلظتها بر روی شاخصهای سرعت و درصد جوانهزنی، وزنتر و خشک ساقهچه و ریشهچه و طول ساقه اثر بهتری داشته است. در آزمایش سوم معلوم گردید که تیمار 4/0 میلیگرم بر میلیلیتر نانوذرات دیاکسید تیتانیوم نسبت به سایر تیمارها موثرتر بود. براساس نتایج حاصل از این آزمایش، پیشنهاد میگردد برای جوانهزنی بهینه بذور توت سیاه، از محیطکشت محتوی نانوذرات دیاکسید تیتانیوم به غلظت 4/0 میلیگرم بر میلیلیتر با شرایط نوری 16 ساعت روشنایی و 8 ساعت تاریکی و همچنین ساکارز به غلظت 15 گرم بر لیتر استفاده شود.

بررسی تنوع اگزون 4 ژن هورمون رشد و ارتباط آن با صفات شیر و چندقلوزایی در بزهای خلخالی
این مطالعه با هدف بررسی تنوع ژنتیکی در ژن هورمون رشد و ارتباط آن با تولید شیر و چندقلوزایی در 120 رأس از بزهای شیری خلخالی انجام گرفت. برای انجام این مطالعه از روش PCR-SSCP جهت شناسایی الگوهای مختلف و توالییابی جایگاه موردنظر استفاده شد. نتایج این مطالعه منجر به شناسایی 4 الگوی مختلف در اگزون 4 ژن هورمون رشد گردید که با بررسی توالیهای حاصل از الگوهای مختلف منجر به شناسایی پنج جهش شد که ایجادکننده 4 هاپلوتیپ I، II، III و IV با فراوانی بهترتیب 24/0، 46/0، 17/0 و 13/0 بود. آنالیز ارتباط اثر هاپلوتیپهای شناسایی شده با صفات تولیدی نشان داد که هاپلوتیپهای مختلف تأثیر معنیداری بر صفات میزان تولید شیر، درصد چربی، درصد پروتئین داشته (p <0.05) درحالیکه تأثیری بر روی صفات درصد لاکتوز و میزان مواد جامد شیر نداشت. همچنین تنوع نوکلئوتیدی و هاپلوتیپی شناسایی شده باعث ایجاد واریانس در صفت چندقلوزایی میشود بهطوریکه حیوانات با هاپلوتیپ II بیشترین میزان چندقلوزایی (159/2) و هاپلوتیپ IV کمترین میزان چندقلوزایی (40/1) را داشتند. نتایج نشان میدهد که هاپلوتیپ II برای صفت چندقلوزایی و هاپلوتیپهای I، II و IV برای صفات تولید شیر و ترکیبات آن میتواند یک نشانگر ژنتیکی مناسب بهشمار رود.

اثر مصرف کود فسفر و نیتروژن بر بیان ژنهای Ta-PHR1 و Ta-PHO2، عملکرد دانه و صفات وابسته در گندم نان
فسفر یکی از مهمترین عناصر مورد نیاز گیاهان میباشد و نقش آن در فرایند تولید و انتقال انرژی است، درنتیجه حفظ و نگهداری فسفر و پاسخ به کمبود مقدار آن در گیاه بهمنظور حفظ و افزایش عملکرد ضروری میباشد. بهمنظور بررسی تأثیر مصرف کود فسفر و نیتروژن بر بیان ژنهای Ta-PHR1 و Ta-PHO2 و افزایش عملکرد دانه در برخی از ژنوتیپهای گندم، آزمایشی بهصورت فاکتوریل-اسپلیت پلات در قالب بلوکهای کامل تصادفی با سه تکرار انجام گرفت. تیمارها شامل دو سطح کود نیتروژن شاهد (0) و 150 کیلوگرم در هکتار و دو سطح کود فسفر شاهد (0) و 100 کیلوگرم در هکتار بهعنوان عامل اصلی و چهار ژنوتیپ گندم (رقم مروارید و لاینهای امید بخش N80-19، T-67-60 و T-65-7-1) بهعنوان عامل فرعی بودند. بهمنظور اندازهگیری عملکرد دانه و اجزای آن در مرحله رسیدگی نمونهبرداری صورت گرفت و برای ارزیابی میزان بیان ژنهای موردمطالعه نمونهبرداری در مرحله پرشدن دانهها صورت گرفت. نتایج نشان داد که بیشترین میزان عملکرد دانه در شرایط مصرف توأم کود نیتروژن و فسفر حاصل گردید و سایر اجزای عملکرد (فاصله میانگره، طول سنبله، طول ساقه، تعداد پنجه بارور، تعداد کل پنجه، تعداد دانه، قطر دانه، طول دانه، وزن هزاردانه) نیز در این تیمار بالاترین مقدار را داشتند. همچنین در بین ژنوتیپها رقم مروارید و لاین N80-19 عملکرد بهتری در هر دو شرایط مصرف و عدممصرف کود داشتند. بیشترین بیان ژن Ta-PHR1در رقم مروارید و لاین T-65-7-1 بود. همچنین مصرف کود فسفر و نیتروژن بر روی تمام صفات مورد بررسی به جز قطر دانه تأثیر مثبت و معنیداری داشت.

بهبود فعالیت آنزیمهای تجزیهکننده سلولز با جهشزایی در قارچ Trichoderma reesei
قارچ Trichoderma reesei یکی از گونههای مهم تولیدکننده آنزیمهای تجزیهکننده سلولز در طبیعت است. در این پژوهش 21 جدایه جهشیافته پرتو گاما از قارچ T. reesei برای تولید آنزیم سلولاز روی محیط محتوی سلولز کلوئیدی غربالگری شد. غلظت پروتئینهای خارج سلولی جدایههای وحشی و جهشیافته با استفاده از روش بردفورد اندازهگیری شد. آویسل، کربوکسی متیل سلولز و کاغذ صافی برای اندازهگیری فعالیت سلولازی استفاده شد. همچنین خلوص و ترکیب پروتئینهای غنی از آنزیم با استفاده از آزمون الکتروفورز ژل پلیآکریلآمید مورد ارزیابی قرار گرفت. بالاترین میزان تولید پروتئین خارج سلولی در جدایههای T. r M7 و T. r M17 مشاهده شد. فعالیت آنزیمهای اندوگلوکاناز، اگزوگلوکاناز و سلولاز کل در جدایه جهشیافته T. r M8 بالاترین مقادیر فعالیت آنزیمی را در بین جدایههای جهشیافته و وحشی نشان داد. اختلاف وزن مولکولی باندهای آنزیمی نشان داد که آنزیمهای EG IV، Cel 1A، Cel 12A، Cel 45A، Cel 3A، Cel 7A، Cel 6A، Cel 5A و Cel 61A سلولز کلوئیدی را بهصورت همافزایی هیدرولیز میکنند. این نتایج نشان میدهد که جهشزایی با پرتو گاما برای دستیابی به جهشیافتههای تریکودرما با تولید بالای آنزیم سلولاز امکانپذیر است.

جداسازی و توالییابی ژن مربوط به آنتیپورتر هیدروژن/ سدیم غشای واکوئلی گیاه هالوفیت (.Kochia scoparia L)
در این پژوهش از گیاه دولپهای و شورزی کوشیا (Kochia scoparia)بهعنوان منبع برای جداسازی ژن استفاده شد. ابتدا با استفاده از پرایمرهای طراحیشده در مناطق حفاظتشدۀ این ژن در گیاهان همخانواده، طول 1605 نوکلئوتیدی و توالی 535 اسیدآمینهای از توالی کدکنندۀ این ژن شناسایی و توالییابی شد. با استفاده از آنالیز BLAST میزان شباهت این توالی با 90%-87% همولوژی در سطح نوکلئوتیدی و 99%-98% در سطح اسیدآمینهای با ژنهایNHX گیاهان خانوادۀ اسفناجیان تعیین شد. آنالیزهای تعیین خصوصیات پروتئینی و ارتباط آن با پروتئینهای دیگر با استفاده از مطالعات بیوانفورماتیکیانجام شد. براساس وجود ساختارهای دوم پروتئینی مشخص شد که در پلیپپتید موردبررسی، 11 مارپیچ آلفا در ناحیه گذرنده از غشا وجود دارد و اسیدآمینههایی با خصوصیت آبگریزی در ناحیه گذرنده از غشا واقع شده است و در ادامه انتهای کربوکسیلی با تعداد کمتری مارپیچ آلفا و میزان آبدوست بودن بالا بهعنوان ناحیۀ سیتوپلاسمی پروتئین موردنظر مطرح میشود. این نتایج نقش دفع سدیم توسط ناقلهای غشای پلاسمایی را برای وجود واکنش مقاومت به شوری در گیاه کوشیا تأیید نمود.

شناسایی گونههای Fusarium در استان لرستان با استفاده از ناحیه بین ژنی فاصلهگذار رونویسیشونده درونی
باتوجه به اهمیت کشت گندمیان در حوزه زاگرس، کشت مکرر و غالب اکثر مزارع این مناطق را گیاهان این خانواده به خود اختصاص میدهند. از میان میکروارگانیسمهای مختلف، گونههای قارچ Fusarium از مهمترین عوامل بیماریزا در این محصولات میباشند. در سالهای 1394 تا 1395 با هدف تعیین گونههای مختلف Fusarium همراه با ریشه و طوقه گندمیان در استان لرستان، از گیاهان زراعی و غیرزراعی مزارع غلات و مراتع مختلف در این استان نمونهبرداری بهعمل آمد. نمونهبرداری از گیاهان دارای علایم پوسیدگی طوقه و ریشه یا تغییر رنگ در این ناحیه انجام و جداسازی عامل قارچی موردنظر با استفاده از محیطهای کشت اختصاصی صورت گرفت. شناسایی جدایهها با استفاده از ریختشناسی و کلیدهای شناسایی و تعیین توالی بخشی از ژنوم شامل ناحیه فاصلهگذار رونویسیشونده درونی (Internal Transcribed Spacer) انجام شد. در این مطالعه 13 گونه از جنس Fusarium شامل F. acuminatum، F. avenaceum، F. crookwellense، F. culmorum، F. equiseti، F. ensiforme، F. falciforme، F. fujikuroi، F. longipes، F. nygamai، F. proliferatum، F. reticulatum، F. scirpi، F. solani و F. tricinctum شناسایی گردید. نتایج نشان داد گونه F. proliferatum بیشترین فراوانی را در این استان دارد. گونه F. ensiforme برای نخستین بار از ایران گزارش میشود و برای فلور ایران جدید است. همچنین این نخستین گزارش از F. falciforme از ریشه گندم در ایران میباشد.
