تولید زیستی نانوذرات نقره با استفاده از قارچ Fusarium oxysporum جدا شده از نخود (Cicer arietinum)

چکیده

یکی از اهداف مهم نانوفناوری توسعه‌ی روش‌های ایمن، سازگار با محیط زیست و کم هزینه برای تولید نانوذرات است و قارچ‌ها به علت ترشح مقادیر زیاد آنزیم و پروسه‌های پایین دست آسان‌تر گزینه‌های مناسبی برای این انتخاب می‌باشند. هدف از این مطالعه تولید خارج سلولی نانوذرات نقره با استفاده از جدایه‌های قارچ Fusarium oxysporum جدا شده از نخود بود. برای این منظور 15 جدایه از قارچ مورد بررسی قرار گرفت. عصاره‌ی سلولی قارچ پس از تیمار با محلول 3-10 مولار نیترات نقره، از بی‌رنگ به قهوه‌ای تغییر یافت که اولین نشانه مبنی بر تشکیل نانوذرات نقره می‌باشد. پس از تولید نانوذرات نقره در مخلوط واکنش، بررسی‌های بیشتر با استفاده از روش‌های اسپکتروفتومتر UV-Vis، XRD و FTIR انجام شد. داده‌های اسپکتروفتومتر وجود پیک در 420 نانومتر را نشان داد که مختص نانوذرات نقره است. طیف‌سنجی هر 24 ساعت یک بار به‌مدت چهار روز ادامه یافت تا در نهایت جدایه F3 که طی این مدت بیش‌ترین جذب و پایداری را از خود نشان داده بود به‌عنوان جدایه‌ی برتر انتخاب و آزمایش‌های تکمیلی روی آن انجام شد. پهن‌شدگی باندها در الگوی XRD بر اندازه‌ی کوچک نانوذرات دلالت داشت. آنالیز FTIR پایداری نانوذرات تولید شده توسط بیومولکول‌ها تا پنج ماه پس از شروع واکنش را تأیید کرد.

In recent years there has been an increased interest to use microorganisms as bionanofactories in nanoparticles synthesis. Secreting high amount of proteins, fungi have advantages over other microorganisms. In the present study 15 Fusarium oxysporum isolates from pea were used for the extracellular biosynthesis of silver nanoparticles. After the addition of AgNo3 solution (final concentration 10-3M), the crude cell filtrate of F. oxysporum changed from colorless to brown color indicating the formation of silver nanoparticles (AgNPs). Aliquots of the cell filtrate were removed and subjected to UV-vis spectroscopy at different time intervals to confirm the formation and stability of AgNPs. Nanoparticles showed an absorption peak at 420nm in UV-visible spectrum corresponding to the Plasmon resonance of AgNPs. Further analysis was carried out by X-ray diffraction (XRD) and Fourier Transform Infrared Spectroscopy (FTIR). Broadening the XRD peaks denote the small size of AgNPs. FTIR analysis confirmed the stability of AgNPs by biomolecules capping agents even 5 months after their production.
