بررسی ارتباط ژن‌های Hd5 و Hd6 با زمان خوشه‌دهی در ارقام محلی و اصلاح شده برنج

چکیده

در این تحقیق ارتباط SNP عملکردی ژن‌های Hd5 و Hd6 با زمان خوشه‌دهی در برنج مورد بررسی قرار گرفت. بدین‌منظور 45 رقم برنج محلی و اصلاح شده در شرایط آب‌وهوایی گرگان مورد ارزیابی فنوتیپی قرار گرفتند و با استفاده از آغازگرهای اختصاصی دو ژن فوق تعیین ژنوتیپ شدند. تجزیه ارتباط در کل جمعیت مورد مطالعه نشان داد که SNP ژن Hd5 8/4 درصد از تغییرات زمان خوشه‌دهی را توجیه نمود و اثر افزایشی آن در جهت دیررسی معادل 6/2 روز برآورد شد. ولی SNP ژن Hd6 5/5 درصد از تغییرات زمان خوشه‌دهی را توجیه نمود و اثر افزایشی آن در جهت زودرسی معادل 4/2- روز برآورد شد. پس از تفکیک جمعیت به دو گروه ارقام اصلاح شده و محلی تجزیه ارتباط نشان داد که اثر SNP ژن Hd5 بر زمان خوشه‌دهی ارقام اصلاح شده غیر‌معنی‌دار بود. اما در گروه ارقام محلی این SNP بخش نسبتاً بالایی (2/16 درصد) از تغییرات زمان خوشه‌دهی را توجیه نمود و اثر افزایشی آن در جهت دیررسی معادل 4/4 روز برآورد شد. اثر SNP ژن Hd6 بر زمان خوشه‌دهی ارقام محلی معنی‌دار نبود، ولی در گروه ارقام اصلاح شده این SNP بخش نسبتاً بالایی (6/17 درصد) از تغییرات زمان خوشه‌دهی را توجیه نمود و اثر افزایشی آن در جهت زودرسی معادل 8/3- روز برآورد گردید. براساس نتایج این تحقیق می‌توان نتیجه‌گیری نمود که اصلاح ارقامی با آلل‌های زودرس‌کننده ژن‌های Hd5 و Hd6 امکان‌پذیر می‌باشد. ‌‌

چکیده [English]

In this research the relationship of functional SNP of Hd5 and Hd6 genes with heading date was investigated in rice. For this purpose, 45 local and improved rice cultivars were phenotypically evaluated in Gorgan condition, and were genotyped using primer pairs specific to the genes. Association analysis in total population showed that Hd5 SNP explained 4.8% of variations in heading date and its additive effect was estimated equal to 2.6 days. However, Hd6 SNP explained equal to 5.5 % of variations in heading date and its additive effect was estimated equal to -2.4 days. After subdividing the population into two local and improved groups association analysis showed that effect of Hd5 SNP on heading date of improved cultivars wasn’t significant. However, in local cultivars, the SNP explained a relatively high portion (16.2%) of variations in heading date and its additive effect was estimated equal to 4.4 days. Effect of Hd6 SNP on heading date wasn’t significant in local cultivars, but in improved cultivars the SNP explained a relatively high portion (17.6%) of variations in heading date and its additive effect was estimated equal to -3.8 days. On the basis of the results of this research it can be concluded that improving cultivars with earliness alleles of both Hd5 and Hd6 genes is possible.
