Optimization of In Vitro Propagation of Alstroemeria by Apical Bud Culture, Callus Induction and Plant Regeneration

Abstract

This research was conducted at two different experiments to optimization of in vitro propagation Alstroemeria. Experiment 1: effect of BAP (0, 0.5, 1, 1.5 and 2 mg/l-1) and IBA (0, 0.1 and 0.2 mg/l-1) on apical buds (0.8-1 cm) grown from young’s shoots tips of Alstroemeria c.v. Fujie and experiment 2: effect of 2,4-D (0, 1, 2, 4, 6 and 8 mg/l-1), picloram (0, 1, 2, 4, 6 and 8 mg/l-1) and BAP (0 and 0.5 mg/l-1) were studied on embryonic callus induction from first seedling leaves in A. ligtu hybrid. The MS medium was used in both experiments. At experiment 1: the highest shoots number was obtained in 1 mg.l-1 BAP+0.1 mg.l-1 IBA (4.27). The results showed that increasing of BAP concentration caused decreasing of shoot and internode length due to apical dominant decreasing via IBA effect. The highest root number (3.25) was obtained in 0.2 mg.l-1 IBA concentration without BAP and the highest rhizome number was recorded at 1 mg/l-1 BAP without IBA (3.5). At experiment 2: maximum of compact embryonic callus (29.13%) was obtained with 2 mg/l-1 picloram and 0.5mg/l-1 BAP, followed by 2 mg/l-1 2,4-D and 0.5 mg/l-1 BAP. To callus proliferation, both types of callus were maintained on MS with 4 mg/l-1 picloram.
