First Report of Eurytoma longavena (Hymenoptera: Eurytomidae) Parasitoid Wasp Species of Oak Gall Forming Wasps from Kermanshah Oak Forests Using Cytochrome Oxidase I. Marker

Abstract

ABSTRACT: During 2018-21 the oak forests Paveh and Ravansar in Kermanshah province was searched for oak gall forming wasps. We found Neuroterus quercusbaccarum as a gall forming wasp species and Eurytoma longavena as its parasitoid wasp species. galls were put in plastic buckets, the location coordinates and date of collection was labeled on buckets.The buckets mouth covered by a white organza net.By daily monitoring of the galls. the exited wasps were collected. The extraction of DNA of specimens were made by use of Sina PureTM DNA kit and quality and quantity of extracted DNA measured. For assessing the DNA quality we used gel electrophoresis with 2% Agarose gel. . PCR was done using three primers and the extracted DNA, For sequencing the PCR product was sent to Topas gene Co. in Tehran. The received COI DNA sequences were edited using Bioedit version 7.1. After which the alignment of our specimen’s sequences with those of Zhang et al., 2013 was made using clustal-W. We sent the edited sequences to Banklit in NCBI and obtained the accession number OL655402 . We used Mega10 software to draw the phylogenetic tree. For checking stable reliabilities of clades of our phylogenetic tree we used bootstrap test with 1000 replications(Fig- 2.). The COI gene for Eurytoma longavena isolated by us had 652 base pairs. We used BLAST in gene bank of NCBI and we found that the COI sequences of our wasp specimen have a 98% overlapping and 98% similarity to the Eurytoma longavena species with code No. MZEUR-0129 and accession number KC685185 collected in Canada (Zhang et al., 2017).
