Molecular Detection of Wheat Dwarf Virus in Cereal Fields of Hamedan Province

Abstract

Wheat dwarf disease which causes by Wheat dwarf virus (WDV), is one of the most important diseases of wheat. This virus is a member of the Mastrevirus genus and Geminiviridae family. WDV is widespread in most parts of Iran. Considering the importance of grain cultivation in Hamedan province, in order to detect of this virus, wheat and barley plants with symptoms of dwarfing and yellowing were sampled from the fields of Malayer, Tuyserkan, Nahavand, Hamedan, Lalejin, Bahar, Asadabad, Famenin, Kabudrahang and Razan counties during growth seasons of 2022 and 2023. Total DNA was extracted from the symptomatic leaves using CTAB buffer. The PCR test was performed using a specific primer pair that amplifies an 847 bp fragment of the WDV genome. According to the results, this virus is present in some sampled areas. The amplified fragments of Famenin, Kabudrahang and Razan isolates were sequenced. Nucleotide sequences of these isolates showed the highest degree of similarity with two isolates of WDV-W species including KU877928 and MH477622 isolates with 99% and 98% similarity, respectively. They have 96% to 98% similarity with other isolates of WDV-W species available in the GenBank. The similarity of these three isolates with the isolates of WDV-B species is between 84% to 86%. This is the first report of wheat dwarf virus (WDV-W) infection in wheat and barley fields in Hamadan province.
