During the examination of papaya plants from the greenhouses around Yazd, Mehriz, Kerman, Bushehr and Chabahar cities in the spring and summer of 1402, samples with yellowness, laceration, small leaves and cupping of leaves, darkening of the petiole and black spot of the fruit symptoms were collected. In order to detect papaya ringspot virus and to investigate the genetic diversity in the collected samples with symptoms, total RNA was extracted from the tissue of leaves with symptoms and without symptoms (negative control) and it was investigated with a pair of general primers of potyviruses (Nib1 and Nib3R) in reverse polymerase chain reaction. The RT-PCR reaction to detect disease-causing viruses in the samples of papaya plants with investigated symptoms showed a positive reaction and led to the amplification of a 350 bp fragment belonging to a part of the nuclear body inclusion protein open reading frame. PCR products obtained were sequenced bilaterally. After NCBI blast of sequences and drawing the phylogenetic tree showed that Iranian PRSV isolates had a high diversity, and in general, most of the examined isolates were very close to Indian and Chinese PRSV isolates.
