بررسی اثرات آلودگی به سرب و کادمیوم بر فعالیت‌های آنزیم‌های آنتی‌اکسیدانی و تحمل به استرس در پنج ژنوتیپ گیاهان دارویی

چکیده
آلودگی فلزات سنگین در خاک‌های کشاورزی به‌ویژه در مناطق صنعتی یک نگرانی فزاینده است که می‌تواند تأثیرات منفی بر رشد و عملکرد گیاهان و سلامت محصولات گیاهی، به‌ویژه گیاهان دارویی، داشته باشد. این مطالعه دوساله به ارزیابی پاسخ‌های آنزیمی و متابولیک پنج ژنوتیپ گیاه دارویی شامل نعناع فلفلی (Mentha piperita)، بابونه (Matricaria chamomilla)، آویشن (Thymus vulgaris)، ریحان (Ocimum basilicum) و رزماری (Rosmarinus officinalis) در معرض سطوح مختلف سرب (Pb) و کادمیوم (Cd) در خاک‌های آلوده به فلزات سنگین در خاک‌های منطقه اراک پرداخته است. آزمایش به‌صورت اسپیلت‌پلات در قالب طرح کاملاً تصادفی با سه تکرار در دو سال متوالی انجام شد که در آن سه سطح سرب (۰، ۲۰۰، ۴۰۰ میلی‌گرم بر کیلوگرم خاک) و سه سطح کادمیوم (۰، ۵، ۱۰ میلی‌گرم بر کیلوگرم خاک) در نظر گرفته شد. در پایان هر فصل رشد، فعالیت آنزیم‌های آنتی‌اکسیدانی شامل سوپراکسید دیسموتاز (SOD)، کاتالاز (CAT)، پراکسیداز (POD)، محتوای مالون‌دی‌آلدئید (MDA) و غلظت کل آلکالوئیدها اندازه‌گیری شد. نتایج نشان داد که قرارگیری در معرض سرب و کادمیوم به‌طور معناداری (در سطح ۱ درصد) بر تمام پارامترهای اندازه‌گیری شده در هر دو سال تأثیر دارد. کادمیوم در سطح ۱۰ میلی‌گرم بر کیلوگرم خاک موجب افزایش قابل‌توجه فعالیت آنزیم‌های آنتی‌اکسیدانی، به‌ویژه سوپراکسید دیسموتاز و کاتالاز، به‌ترتیب ۴۲ درصد و ۳۷ درصد در سال اول و ۴۵ درصد و ۴۰ درصد در سال دوم شد. در عین حال، محتوای مالون‌دی‌آلدئید به‌ترتیب ۵۵ درصد و ۵۸ درصد افزایش یافت. ترکیب Pb400 + Cd10 بالاترین میزان پراکسیداسیون لپید را در سال اول (۶۸ درصد) و سال دوم (۷۱ درصد) نشان داد. در شرایط تنش متوسط، میزان آلکالوئیدها ۱۸-۲۰ درصد افزایش یافت، در حالی که تنش شدید به‌طور معناداری سطح آلکالوئیدها را به میزان 23-۲۰ درصد کاهش داد. در پاسخ‌های وابسته به گونه، رزماری قوی‌ترین سیستم دفاعی آنتی‌اکسیدانی و ریحان بیشترین میزان پراکسیداسیون لپید را از خود نشان داد. این یافته‌ها بر لزوم پایش بلندمدت آلودگی فلزات سنگین در خاک‌های کشاورزی و تأثیرات آن بر کیفیت و ایمنی گیاهان دارویی تأکید می‌کند و می‌تواند به‌عنوان مرجعی برای انتخاب ژنوتیپ‌های مقاوم به فلزات سنگین در برنامه‌های اصلاح گیاهان دارویی مورداستفاده قرار گیرد.

Investigation of the Effects of Lead and Cadmium Pollution on Enzymatic Activities and Stress Tolerance in Five Genotypes of Medicinal Plants

Heavy metal pollution in agricultural soils, particularly in industrial areas, is a growing concern that can negatively affect plant growth, performance, and the health of herbal products. This two-year study evaluated the enzymatic and metabolic responses of five medicinal plant genotypes—Peppermint (Mentha piperita), Chamomile (Matricaria chamomilla), Thyme (Thymus vulgaris), Basil (Ocimum basilicum), and Rosemary (Rosmarinus officinalis)—exposed to various levels of Lead (Pb) and Cadmium (Cd) in heavy metal-contaminated soils of the Arak region.The split-plot factorial experiment was conducted in a completely randomized design with three replications over two consecutive years. Three levels of Lead (0, 200, 400 mg/kg soil) and three levels of Cadmium (0, 5, 10 mg/kg) were tested. At the end of each growing season, the activity of antioxidant enzymes, including Superoxide Dismutase (SOD), Catalase (CAT), Peroxidase (POD), the content of Malondialdehyde (MDA), and the total concentration of alkaloids, were measured. The results showed that exposure to Lead and Cadmium significantly (at the 1% level) affected all measured parameters in both years. Cadmium at a concentration of 10 mg/kg soil led to a significant increase in the activity of antioxidant enzymes, particularly SOD and CAT, by 42% and 37% in the first year, and 45% and 40% in the second year, respectively. Concurrently, MDA content increased by 55% and 58%, respectively. The combination of Pb400+Cd10 showed the highest level of lipid peroxidation at 68% in the first year and 71% in the second year. Under moderate stress, alkaloid levels increased by 18-20%, while severe stress significantly reduced alkaloid levels by 23-25%. In species-dependent responses, Rosemary (Rosmarinus officinalis) exhibited the strongest antioxidant defense system, while Basil (Ocimum basilicum) showed the highest level of lipid peroxidation. These findings emphasize the necessity of long-term monitoring of heavy metal contamination in agricultural soils and its impact on the quality and safety of medicinal plants, providing a reference for selecting heavy metal-tolerant genotypes in medicinal plant breeding programs.
