تکثیر درون شیشه‏ ای انار رقم رباب نیریز با استفاده از قطعات گره‏ای ساقه

چکیده

در این تحقیق، یک روش سریع و کارآمد برای تکثیر درون­شیشه‏ای انار رقم رباب نیریز شرح داده می‏شود. اثر دو محیط­کشت WPM و MS، و تنظیم­کننده‏های رشد مختلف در ریزازدیادی این رقم بررسی شد. برای مرحله پرآوری، محیط­کشت دارای غلظت‏های مختلف کینتین (3/2، 7/4، 2/9 و 4/18 میکرومولار) همراه با 54/0 میکرومولار نفتالین استیک­اسید استفاده گردید. از نظر پرآوری، محیط­کشت WPM در مقایسه با MS کارآمدتر تشخیص داده شد. بهترین غلظت کینتین 2/9 میکرومولار بود که منجر به بیش­ترین طول شاخساره (93/3 سانتی­متر)، تعداد برگ (71/10) و تعداد گره (4) در ریزنمونه‏ها شد. محیط نصف غلظت WPM حاوی 4/5 میکرومولار NAA مؤثرترین محیط برای ریشه­زایی شاخساره‏ها بود. گیاهان ریشه­دار شده به­طور موفقیت­آمیزی سازگاری تدریجی شده و به خاک منتقل گردیدند. گیاهان ریز ازدیادی یافته از نظر مرفولوژیکی شبیه به هم بوده و صفات رویشی مشابه با گیاهان مادری نشان دادند.

چکیده [English]

In this study, an efficient in vitro propagation method is described for Punica granatum cultivar ‘Rabab Neyriz’. The influence of two basal medium, WPM and MS, and different plant growth regulators was investigated on micropropagation of this cultivar. For proliferation stage, media supplemented with different concentrations (2.3, 4.7, 9.2 and 18.4 μM) of kinetin along with 0.54 μM NAA was used. On base of proliferation, WPM proved to be more efficient medium compared to MS. The best concentration of kinetin was 9.2 μM resulting in the highest shoot length (3.93 cm), leaf number (10.71) and node number (4). Half-strength WPM medium supplemented with 5.4 μM NAA was most effective for rooting of shoots. Rooted plantlets were successfully acclimatized and transferred into soil. The micropropagated plants were morphologically uniform and exhibited similar growth characteristics and vegetative morphology to the mother plants.
